The host laboratory, lead by Nicolas Mirouze (ORCID: 0000-0002-1038-0495) has an extensive experience in the investigation of natural competence for genetic transfoormation in several model organisms. The main originality of this project resides in our choice to study a new model organism. Indeed, taking advantage of the knowledge accumulated in 'old' model organisms and of an important set of preliminary data, we propose to investigate Horizontal Gene Transfer (HGT) through competence and genetic transformation in S. aureus, an important human pathogen often associated to multi-antibiotic resistances. Identifying the signal leading to competence development, characterizing the regulation pathways transmitting this signal as well as deciphering the actors responsible for DNA binding and transport during transformation should allow us to understand how this bacterium evolved to become a major human pathogen. In order to investigate all these aspects we propose to conduct high- throughput and unbiased approaches (Transposon-sequencing, Yeast-Two-Hybrid) as well as to verify hypotheses formulated thanks to our preliminary data (through genetics, RNA-sequencing, microsocpy...). Finally, we propose to verify the results obtained in vivo. To do so, a S. aureus strain collection as well as infectious modeles (mice) will be available through our collaborators.

The postdoctoral researcher will be involved in one of the host laboratory research project which aims to investigate the development of natural competence (an environmental adaptation leading to horizontal gene transfer through genetic transformation) in the human pathogen Staphylococcus aureus. The candidate, helped by the host laboratory will apply to the European Commission Marie Sklodowska-Curie Postdoctoral Fellowship (HORIZON- MSCA-2022-PF-01).

The host laboratory is part of the Microbiology department within the Institute for Integrative Biology of the Cell (I2BC, https: // www. i2bc.paris- saclay.fr/). The host institute (CNRS/CEA/UNiversité Paris-Saclay) is divided in 5 scientific departments (700 researchers), linked to numerous technological platforms (microscopy, high throughput sequencing, biochemistry...).
